Antioxidant level of uterus
A 1 g uterine sample was taken from -20 °C and mixed with 9 ml of phosphate-buffered saline (PBS, 0.01 mol L-1 170 , PH = 7.2-7.4). After 5 min of homogenization with a 5000-rpm high-speed homogenizer, and 10% tissue homogenate was prepared. This was centrifugated at 3500 rpm for 10 min, and the supernatant was taken for the detection of the antioxidant index. Kits for measuring the total protein(Code: JA10245), total antioxidant capacity (T-AOC) (Code: J-13106), superoxide dismutase (SOD) (Code: J-50065), glutathione peroxidase (GPx) (Code: J-53063), catalase (CAT) (Code: J-52619), glutathione S-transferase (GST) (Code: J-52618) enzyme activity and reduced glutathione (GSH) (Code:J-53087), malondialdehyde (MDA) (Code: J-58063), and hydrogen peroxide (H2O2) (Code: J-35958) content in tissues were purchased from Shanghai Jinma Laboratory Equipment Co., Ltd. (Jinma, Shanghai, China). All steps were performed in strict accordance with the kits’ instructions. The measured concentrations were based on the total protein mass of the uterus.

Evaluation of antioxidant enzyme activity and the concentration of nitric oxide Superoxide dismutase (SOD), glutathione peroxidase(GSH-Px) and catalase (CAT) enzyme activities in the milk, plasma and placenta were determined using commercially available kits (Nanjing Jiancheng Bioengineering Institute, Nanjing, China) in accordance with our previous study. The results of the measurements were expressed as U/mL in plasma and milk and as U/mg protein in placenta. Lipid peroxidation in the plasma,milk and placenta was determined by measuring the amounts of malondialdehyde (MDA) through the thiobarbituric acid method using commercially available kits (Nanjing Jiancheng Bioengineering Institute, Nanjing,China). The results of the measurements were expressed as nmol/mL in plasma and as nmol/mg protein in placenta. Nitric oxide (NO) and inducible NOS (iNOS) in the plasma and placenta of sows were determined using assay kits obtained from Jiancheng Biochemistry (Nanjing, China). The colostrum was analyzed for lactose,protein, fat, and total solids with a fully automatic milk analyzer (Milko ScanTM FT+ Analyzer, Foss). The milk samples were analyzed for immunoglobulin G (IgG), immunoglobulin A (IgA), and immunoglobulin M (IgM) using immunoglobulin-specific kits (Jinma Biological Engineering Co., Ltd., Shanghai, China).

To determine the antioxidant indexes, a 10 % liver/PBS (0.01 mol/L, pH = 7.2–7.4) homogenate was first prepared. The liver SOD, GSHPx, GST, CAT, GSH and MDA levels were determined using ELISA kits,and T-AOC and H2O2 levels were determined by colorimetry. All the kits were purchased from Shanghai Jinma Laboratory Equipment Co.,Ltd. (Jinma, Shanghai, China), and each indicator was detected strictly according to the kit’s specifications.
